T cell receptor diversity evaluation to predict patient response to Ipilimumab in metastatic melanoma
نویسندگان
چکیده
Background Ipilimumab blocks the immunologic checkpoint cytotoxic T lymphocyte antigen-4 (CTLA-4) and improves overall survival in patients with metastatic melanoma. Since only a subset of patients achieves benefit from ipilimumab, biomarkers that can predict patient outcome are needed. CTLA-4 blockade has been shown to affect the peripheral T cell receptor (TCR) pool, but whether diversity of the peripheral TCR repertoire is relevant as a biomarker for ipilimumab remains unknown.

Ipilimumab: A First-in-Class T-Cell Potentiator for Metastatic Melanoma
Ipilimumab, a fully human anti-cytotoxic T-lymphocyte antigen-4 monoclonal antibody that potentiates antitumor T-cell responses, has demonstrated improved survival in previously treated and treatment-naïve patients with unresectable stage III/IV melanoma. Survival benefit has also been shown in diverse patient populations, including those with brain metastases. In 2011, ipilimumab (3 mg/kg ever...
متن کاملPeripheral T cell receptor diversity is associated with clinical outcomes following ipilimumab treatment in metastatic melanoma
BACKGROUND Ipilimumab improves overall survival in a subset of patients with metastatic melanoma. Peripheral blood T cell receptor (TCR) repertoire diversity has been associated with favorable outcomes in patients with cancer, but its relevance as a biomarker for ipilimumab outcomes remains unknown. FINDINGS In this pilot study, we analyzed the pre-treatment peripheral blood TCR repertoire in...
متن کاملCutaneous melanoma: new advances in treatment*
Cutaneous melanoma is a challenge to treat. Over the last 30 years, no drug or combination of drugs demonstrated significant impact to improve patient survival. From 1995 to 2000, the use of cytokines such as interferon and interleukin become treatment options. In 2011, new drugs were approved by the U.S. Food and Drug Administration, including peginterferon alfa-2b for patients with stage III ...
متن کاملMetastatic melanoma response to combination therapy with ipilimumab and vemurafenib.
Combination therapies for the treatment of metastatic melanoma are a matter of debate nowadays. We report on a stage IV metastatic melanoma patient with the BRAF V600 mutation and a large tumor burden initially treated with two cycles of ipilimumab. Due to dramatic disease progression, demonstrated on interim 18F-FDG PET/CT, vemurafenib was added in the patient's therapeutical scheme. After com...
متن کاملLate Onset Ipilimumab-Induced Pericarditis and Pericardial Effusion: A Rare but Life Threatening Complication
Metastatic cutaneous melanoma has poor prognosis with 2-year survival rate of 10-20%. Melanoma cells express various antigens including gp100, melanoma antigen recognized by T cells 1 (MART-1), and tyrosinase, which can induce immune-mediated anticancer response via T cell activation. Cytotoxic T-lymphocyte associated antigen-4 (CTLA-4) is an immune check point molecule that negatively regulate...
